Effects of Different Concentration of Vitreous on Cultured Bovine Retinal Pigment Epithelial Cells: In Vitro Study

Abstract

In recent years it has been clear that the vitreous gel which is a specialized extracellular matrix plays an active role in regulating behavior of RPE cells. Although the factors controlling RPE cells behavior are poorly understood yet. We treated cultured bovine RPE cells with different concentrations of vitreous and investigated its effects on cell proliferation and growth. We also performed polyacrylamide gel electrophoresis to see the possibility of change in vitreal component by filtration. RPE cells were counted with coulter counter. Major findings obtained from this study are as follows: 1. The whole vitreous at different concentrations(10%, 20% and 40%) stimulated the proliferation of bovine RPE cells significantly(P<0.05) with the maximum degree at 20% concentration. Vitreous treated bovine RPE cells showed no morphology growth pattern. 2. Treatment with vitreous failed to induce any change in microscopic morphology or growth pattern of RPE cells. 3. Electrophoresis indicated no difference in vitreous component between preand post-filtration and showed maximal band at protein with molecular weight of 66KD.
