Experimental Study on the Toxicity of Carboplatin Infused into Proper Hepatic Artery of Rabbits

BACKGROUND/AIMS: The aim of this study was to determine hepatic and renal toxicity of carboplatin infused into the hepatic artery of rabbits through transfemoral catheterization.
METHODS
Twenty-five rabbits were evenly divided into five groups. In control group, contrast media was infused. In four experimental groups, 0.5, 1.0, 5.0, and 10.0 mg/kg of carboplatin were infused into the hepatic artery, respectively. We evaluated residual plasma platinum concentration, red blood cell (RBC) and white blood cell (WBC) counts, body and liver weights, and histologic findings of liver and kidney to determine toxicity of carboplatin infused into the hepatic artery.
RESULTS
The mean concentration of residual plasma platinum was 44.8% at 4 days after the infusion. The RBC and WBC counts by the 20th day after infusion of carboplatin were within normal Iimits. The body weights were increased only 0.09+-0.14 kg by the 30th day after infusion of carboplatin compared with 0.44+-0.15 kg in control group. The liver weights were increased less in the experimental group than in the control group. The histologic findings of the liver and kidney specimens revealed multifocal inflammatory cell infiltration which increased in correlation to the dose of carboplatin.
CONCLUSIONS
The 10 mg/kg of carboplatin is safe dose to infuse into the hepatic artery without serious toxicity.
