Evaluation of ADVIA Apolipoprotein Reagents on the ADVIA 1650(R) Chemistry Analyzer

Abstract

BACKGROUND: Apolipoprotein A-I (apo A-I) and apolipoprotein B (apo B) are the risk factors for cardiovascular diseases. The apo B/apo A-I ratio has also been superior to the LDL-C/HDL-C and TC/HDL-C ratios in predicting cardiac risk.
METHODS
We evaluated ADVIA Apo A-I and Apo B Reagents (Bayer HealthCare Ltd., USA) using ADVIA 1650(R) (Bayer HealthCare Ltd.), automated chemistry analyzer. We analyzed the precision, linearity, carryover, correlation with BN II (Dade Behring Inc., USA), and reference ranges according to CLSI guidelines.
RESULTS
The coefficients of variation (CV) of both within-run and total precisions were below 2.5% for apo A-I and apo B, and good linearity was observed for both within measurement ranges. Coefficients of correlation were also good (r>0.98), but the mean differences between ADVIA 1650 and BN II was 28.9 mg/dL (17.1%) for apo A-I (P>0.05) which became larger as the apo A-I concentrations were higher.
CONCLUSIONS
ADVIA reagents for apo A-I and apo B showed good precision, linearity, and carryover. The significant difference of apo A-I value between two analyzers could be largely explained by the high apo A-I value assigned to the ADVIA calibrator. We conclude that the reagents are appropriate for routine clinical use.
