Generation of Leukemic Dendritic Cells from Patients with Acute Myelogenous Leukemia

We investigated the generation of dendritic cells (DCs) from acute myelogenous leukemia (AML) cells and their potent ability stimulating leukemic specific cytolytic activity. To generate leukemic DCs, 1 x 107 mononuclear cells isolated from 12 AML patients were cultured in a medium containing GM-CSF, IL-4, TNF-alpha, and SCF for 8-10 days. Nine out of the 12 AML samples showed leukemic DCs generation. The median number of leukemic DCs harvested was 6 x 106 cells, ranging from 2 x 106 cells to 1.1 x 107 cells. The leukemic DCs exhibited high expression of CD11c, HLA-DR, and CD54, and intermediate expression of CD86 and CD1a. However, there was no expression of CD83, CD40, and CD14 in the leukemic DCs. The leukemic DCs were capable of stimulating allogeneic T-cells and cytotoxic activity to autologous leukemic cells. These results suggest that AML cells could be used to generate leukemic DC and the leukemic DC could induce anti-leukemic cytotoxicity.
