Diversity of HLA-A26 Alleles and Haplotypes in Koreans

Abstract
- BACKGROUND
HLA-A26 is an antigen belonging to the HLA-A10 crossreacting group and shows a gene frequency of 6.7% in Koreans. A26 is encoded by at least 17 distinct alleles. In this study, the distribution of HLA-A26 alleles and their association by HLA-B, -Cw and -DRB1 alleles were investi-gated in the Korean population. METHODS: A total of 453 healthy unrelated Koreans including 215 parents from 108 families were studied. For all samples, HLA-A, B serological typing, HLA-Cw low resolution typing using the polymerase chain reaction (PCR)-sequence specific oligonucleotide (SSO) method and HLA-DRB1 genotyping using the PCR-single strand conformation polymorphism (SSCP) method were performed. All of the 62 samples (13.7%) typed as HLA-A26 by serology were analyzed for A26 alleles using the PCR-low ionic strength (LIS)-SSCP method. RESULTS: Three HLA-A26 alleles, A*2601 (gene frequency 3.8%), A*2602 (2.3%) and A*2603 (1.1%), were observed in the Korean population. Significant associations (P<0.05) were observed for A*2601 with Cw*03, B27, B35, B61, and DRB1*1201; A*2602 with Cw*03, Cw*1402, DRB1*0901, and DRB1* 1501; and A*2603 with Cw*03. CONCLUSIONS: The Korean population shows a heterogeneity in the distribution of HLA-A26 alleles and related haplotypes, and thus the A26 subtype mismatch can occur between serologically-matched donor-recipient pairs in unrelated marrow transplantations.
