Analysis of CYP2E1 and GSTM1 Polymorphisms for the Determination of Genetic Susceptibility to Korean Patients with Colon Cancer

BACKGROUND/AIMS: Genetic susceptibility to chemical carcinogens is one of the most important host factors in human cancers. This study was undertaken to reveal the association of the polymorphism of the cytochrome P450 (CYP2El/PstI, and CYP2E1/DraI) and the glutathion S-transferases isozymes (GSTM1) with the genetic susceptibility of colon cancer development in Korean.
METHODS
In this study, 57 patients with colon cancer and 145 controls were analysed using polymerase chain reaction- restriction fragment polymorphisms (PCR-RFLPs).
RESULTS
In the analysis of CYP2E1 polymorphisms, the genotypes of the controls and patients with colon cancer are well fitted to the HardyWcinberg equilibrium. However, significant differences were noted in the genotype distributions of' CYP2E1/DraI polymorphisms between the controls and the patients with colon cancer (p=0.0068). The C/D genotypes were significantly represented in young patients (<50 years old)(p=0.0466) and in smoking patients (p=0.0011). In the analysis of CYP2E1/PstI polymorphism, there were no statistic differences in the distribution of genotypes between the patients and the controls. However, the cl/c2 genotypes were significantly represented in young age group (<50 years old)(p=0.0352).
