Altered Expression of Genes in a Human Urinary Bladder Transitional Cell Carcinoma Cell Line of Low-metastatic Potential and Its Subclone of High-metastatic Potential

Invasion and metastasis are biologic hallmarks of malignant tumors. For tumor cells to break loose from a primary mass, enter blood vessels or lymphatics, and produce a secondary growth at a distant site, they must go through a series of steps. The purpose of this study is to isolate genes of altered expression in a pair of transitional cell carcinoma cell line of the human bladder with low-metastatic potential and that of high-metastatic potential using representational difference analysis. The 253J is a transitional cell carcinoma cell line with low-metastatic potential, and the 253JB-V is a subclone of 253J with high-metastatic potential. The subtraction was done in two directions. For forward subtraction ds cDNA of 253J was used as a tester and that of 253J B-V as a driver, and for reverse subtraction ds cDNA of 253J B-V was used as a tester and that of 253J as a driver, respectively. The forward subtraction identified interferon induced trans- membrane protein 3 (IFITM3) that is strongly expressed in 253J and weakly expressed in 253J B-V in northern blot. The reverse subtraction designated VGF that is strongly expressed in 253J B-V and weakly expressed in 253J both in northern blot and western blot. Although in vivo and in vitro selection cannot be ruled out as contributing to the metastatic phenotype, the up-regulation of VGF expression seem to be involved in the high-metastatic phenotype of 253J B-V cell line. And up-regulation of IFITM3 seem to be involved in the suppression of metastatic phenotype of 253J cell line.
