Nuclear Factors Bind to the Specific Sites in Promoter Region of Human YB-1 Gene

Abstract

This study was designed to identify the presence of the nuclear factors binding to promoter region of human YB-1 gene and determine their specific binding sites. Nuclear extracts were prepared from Chang liver, Hep G2 and CT-26 cells and electrophoretic mobility shift assay was performed to determine the specific binding sites. The nuclear factors isolated from three cell lines commonly bind to the C8 (+96~+125) site in promoter region of YB-1 gene. The nuclear factors prefer binding to double-stranded DNA rather than binding to single-stranded DNA. These data suggest that C8 is the important binding site for the nuclear factors presumably regulating the transcription of human YB-1 gene.
