Construction of Genetically Defined aroA Deletion Mutants of Shigella sonnei KNIB104S

Abstract

In order to develop vaccine candidates against Shigella sonnei KNIH104S, which was isolated in Korea, we have constructed genetically defined aroA deletion mutants using a suicide vector system. They can grow on a chemically defined minimal medium (M9) supplemented with aromatic compounds, but can not grow on a minimal medium without supplements. It was confirmed that the aroA gene had a deletion by PCR technique and Southern hybridization analysis.
