An Experimental Study on Isoniazid plus Thiacetazone Chemotherapy in Experimental Mice Tuberculosis

Abstract

This study was designed to observe the effect of Thiacetazone in experimental mice tuberculosis. Experimental animals were divided into two groups, namely Isoniazid alone and Isosciazid plus Thiacetazone combined group. Body weight, mortality and macroscopic finding of the lungs were observed, and viable count and sensitivity test to Isoniazid and Thiacertazone were carried out on 30 th, 90th, and 180th day. The results were as follows: 1) The change of body weight showed no remarkable difference between Isoniazid alone and Isoniazid plus Thiacetazone combined group. 2) No mortality was observed in Isoniazid alone, but 24% was observed isoniazid plus Thiancetazone combined group which is considered probably due to toxicity of Thiacetazone. 3) Aoki's macroscopic finding of the lungs were not significantly different between both groups. 4) The viable counts of the lungs were averaged 5.0220 on 30th day, 3.8585 on 90th day and 2.5230 on 180th day in Isoniazid alone group, In Isoniazid plus Thiacetazone combined group, the viable counts were averaged 5.5563 on 30th day, 3.4761 on 90th day and 2.1405 on 180th day. These findings may indicate no remarkable differences between both groups. 5) In Isoniazid alone all of 5 mice were sensitive on 30th day. 3 of 5 were resistant to 0.1-0.5 mcg/ml on 90th day and 4 of 5were resistant to 1.0-5.0 mcg/ml on 180th day, on the other hand in Isoniazid plus Thiacetazone combined group, all of 5mice were sensitive on 30th day, 1 of 4 were resistant to 0.1 mcg/ml on 90th day and 2 of 5 were resistant to 0.1 mcg/ml, and 3 of 5 were resistant to 5 mcg/ml on 180th day. 6) The results of Thiacetazone sensitivity test were so variable, that we can not conclude whether sensitive or resistant. 7) The above results may indicate that there were no significant differences in therapeutic effect and production of resistant strains to Isoniazid between two groups.
