Immunoblotting Analysis on Antigenic Components of Crude Cell Extract (TE) from Mycobacterium tuberculosis with Sera from Tuberculosis Patients

Abstract

This study was aimed to identify antigenic components of Mycobacteηum tuberculosis having relative specificity and sensitivity for the serodiagnosis of pulmonary tuberculosis. Sonicated cell extract (TE) of M. tuberculosis was used as an antigen with which sera from tuberculous or non-tuberculous patients and from normal persons were reacted. Three antigenic components (135KD, 92KD, 28KD) were found to be useful for the diagnosis of patients infected with mycobacterial species because they reacted only with sera from patients with pulmonary tuberculosis Two antigenic components (38KD, 24KD) reacted with sera from tuberculosis patients which had been absorbed with sonicated nontuberculous mycobacterial cell extracts. 38KD-protein of them was found to be more useful than 24KD-protein in discrimination of tuberculosis from non-tuberculosis patients because antibodies to the component were detected in sera from 9 of 13 tuberculosis patients while 24KD-protein was reacted with sera from 3 of 13 tuberculosis patients. Four antigenic components (41KD, 33KD, 29KD, 28KD) weakly reacted or did not react at all with sera from tuberculosis patients that had been collected after completion of successful treatment. Especially 28KD-protein was found to be useful in discrimination terated persons from unterated cases fairly well because it did not react with sera from 7 of 10 patients that had been collected after completion of successful treatment.
