Evaluation on Usefulness of L-pyrrolidonyl-beta-naphthylamide (PYR) Test and NaCl-Esculin Hydrolysis Test for the Rapid Presumptive Identification of Enterococci

BACKGROUND: The rapid and accurate identification of enterococci can provide clinician's decision making of antimicrobial therapy because enterococci are usually multiresistant to commonly used antimicrobial agents and antimicrobial resistance patterns are different according to enterococcal species. So, we evaluated the usefulness of L-pyrro-lidonyl-beta-naphthylamide (PYR) test and NaCl-esculin hydrolysis test for the presumptive identification of enterococci.
METHODS
One hundred eighty-eight enterococci, 227beta-hemolytic streptococci, 10 non-beta-hemolytic streptococci, one Aerococcus viridans, two Micrococcus luteus, and three Lactococcus garvieae were simultaneously tested with PYR and NaCl-esculin hydrolysis test.
RESULTS
All of enterococci and S. pyogenes were positive in the PYR test, while all of beta-hemolytic streptococci other than S. pyogenes and non-beta-hemolytic streptococci were negative in the PYR test. The rates which showed positive reaction of NaCl-esculin hydrolysis test by organisms were as follows: enterococci ; 99.5%, both of group A and B streptococci ; 0%, each of group C, G, and F streptococci ; were 8.3%, 45.5%, and 6.6%, respectively.
CONCLUSIONS
PYR test in combination with NaCl-esculin hydrolysis test are rapid and accurate presumptive identification system for enterococci and S. pyogenes.
