The Effect of 17-Beta Estradiol Local Delivery on Endothelial Cell Function in Porcine Coronary Model

The aim of this study is to investigate the consequences of local delivery of 17-beta estradiol (estradiol) on endothelial function and vasomotor reactivity. Sixteen coronary arteries of eight pigs were randomly assigned to two groups: control vehicle delivery (group I, n=8) and estradiol delivery (group II, n=8). Endothelial function was assessed at 28 days after the local delivery. After achieving steady-state, KCl-induced baseline tension ratio was not different between the two groups. The sodium nitroprusside- induced endothelial independent relaxation ratio and endothelium-derived relaxing factor (EDRF) induced relaxation ratio were not different between the two groups at 4-week (I: 0.94+/-0.43, II: 0.98+/-0.58, p=0.366, I: 0.98+/-0.58, II: 1.12+/-0.30, p=0.321, respectively). However, endothelium-derived hyperpolarizing factor (EDHF) induced relaxation ratio was significantly lower in group I compared with group II at 4 weeks (I: 0.68+/-0.31, II: 1.17+/-0.26, p=0.035). In conclusion, local estradiol local delivery prevents endothelial dysfunction by endothelial-derived hyperpolarizing factor in a porcine coronary model.
