HLA-DR Typing with PEL-FREEZ(R) DR4 TEST-SSP UNITRAY(TM)

Abstract

BACKGROUND: PEL-FREEZ(R) DR4 TEST-SSP UNITRAY(TM) (PEL-FREEZ) was recently introduced in Korea, which utilized the technique of sequence specific primer-polymerase chain reaction for HLA-DR typing.
METHODS
The performance of PEL-FREEZ was compared with that of Dynal RELL(TM) SSO HLA-DRB Test Kit (Dynal) using 8 samples among which there were 4 samples with ambiguous results and another 4 samples assigned of their HLA-DR with certainty by Dynal. 100 clinical samples were typed of HLA-DR by PEL-FREEZ.
RESULTS
HLA-DR results of 4 samples assigned by Dynal were identical with those of PEL-FREEZ. HLA-DR of another 4 samples could be assigned with certainty by PEL-FREEZ. HLA-DR of another 4 samples could be assigned with certainty by PEL-FREEZ. PEL-PREEZ was able to clearly distinguish DR15 from DR16, DR13 from DR14, and DR13 from DR11, HLA-DR antigens of 100 clinical samples were assigned with certainty by PEL-FREEZ. There were 2 cases which were not assigned of their HLA-DR due to false positive bands and needed repeated tests.
CONCLUSIONS
PEL-FREEZ(R) DR4 TEST-SSP UNITRAYTM kit was easy and unambiguous to interpret the result of HLA-DR. This will be very good to use in laboratories in which only several samples are being typed in a run.
