Growth of Staphylococcus Aureus in Tissue Culture of Monkey and Porcine Kidney Epithelial Cells

It is well known that pathogenic Staphylococcus aureus produces various kinds of pathologic lesion in animal, however, it is difficult to observe growth sites of the staphylococcus and cellular lesion in kidney tissues in vivo. This experiment was designed to study the growth of Staphylococcus aureus in cultures of monkey and porcine kidney epithelial cells in vitro. The results of observation are summarized as follows. 1. Multiplication of Staphylococcus aureus was better in kidney epithelial cell culture tubes than medium 199 containing 2% calf serum. Maximum number of the bacteria was observed on day 2 to 4 days after inoculation of staphylococcus into the cell culture tubes and reproduction of the staphylococcus at the peak has reached one billion times inoculation dose. 2. Monkey kidney epithelial cells was more susceptible to Staphylococcus aureus i.e. complete cell destruction were observed in 48 hours than porcine kidney epithelial cells which showed cell destruction 4 days after inoculation. 3. Some relationship between growth of Staphylococcus aureus and cytopathologic changes of kidney epithelial cells was observed in vitro.
