Comparison of three supravital stains and acridine orange test in examination of human semen

Abstract

Twelve human semen specimens were used to compare the reliability of the eosin, trypan blue, and hoechst supravital tests. The acridine orange test was also carried out to investigate whether the score of this test has a relation to the results of supravital testing. All specimens were tested in duplicate. The percentages of live sperm found by eosin, trypan blue, and Hoechst staining were 50%-88%, 46%-89%, and 48%-87%, respectively. The differences are not statistically significant. The sperm motility scores had no relation to the supravital staining results, as many sperm were viable even though motility was 0.
