Separation of Tissue proteins in Transitional Cell Carcinoma of Bladder by Two-dimensional Electrophoresis

The heterogeneous mixture of proteins present in high grade, invasive transitional cell carcinoma of bladder from thirteen patients were analysed by two-dimensional gel electrophoresis in comparison with normal bladder tissue. We carried out one-dimensional gel electrophoresis to all tissue specimens for screening and three different staining methods were applied to discriminate whether these separated proteins were glycosylated or not. Our results show that many polypeptides found with different degrees of frequency in bladder carcinomas were abscent or diminished in amount of protein from non-tumorous bladder tissues. Among these polypeptides, proteins 41/6.7-7.0, 38.5/7.4-8.0, 31/8.0 and 29.5/5.2-5.3(molecular weight/isoelectric point) were detected constantly with all three different staining methods. We suggest that these proteins are possible tumor-associated glycoproteins in high grade, invasive transitional cell carcinoma of bladder. Further studies are required to verify the specificity before they can be considered for bladder tumor-specific proteins.
