Effect of Staining Method on Spermatozoa Morphology

Abstract

A study was carried out to analyze how the results of sperm morphology assessment are influenced by the various staining methods recommended for this purpose: the eosin-nigrosin(E-N), Papanicolaou(Pap), and Diff-Quick(D-Q) methods. Semen analysis and detailed classification of the abnormal sperm morphology was performed according to the WHO standard. The mean percentages of normal morphology were 68.8+/-1.2%(E-N), 76.0+/-1.3%(PAP), and 77.4%(D-Q). Analysis by correlation coefficient showed that specimens with oval, large, small, precursor, or coiled abnormal morphologies produced significantly different results with eosinnigrosin staining than with the other methods. The Pap and D-Q methods are recommended for strict evaluation of spermatozoa morphology in andrology laboratories.
